-pGLO DNA bacteria will be gram- negative bacteria.  +pGLO DNA bacteria will be gram positive bacteria.

+pGLO DNA bacteria will have a thick peptidoglycan layer and no outer lipid membrane.

-pGLO DNA bacteria will have a thin peptidoglycan layer and also an outer lipid membrane will be present.

The basic difference between the two will be the presence or absence of the outer lipid membrane.
